Molecular genetic mechanisms of vertebrate retina during ontogeny

N.K.Koltsova, Moscow, Russia The retina of an eye is formed from primary neuroectoderm which is localized in a forward part of a nervous plate, further formation of a nervous tube and division of a field of an eye into two rudiments, form pair optical везикул. Formation of a two-layer optical bowl needs invaginatsiya optical vesicles. Its back layer forms pigmentary retina epiteliya, and its forward - a neuroretina. During a differentiation are formed 6 main cellular types of cages: amakrinovy, bipolar, horizontal, fotoretseptorny, gangliozny and cages of the Myullerovsky glia. A retina of vertebrata, at molecular level, is characterized by activation of several transkriptsionny factors (TF) of families: Rax, Pax, Six, Otx, Chx and др. These TF can operate as transkriptsionny activators and/or repressor. Mutations of genes of regulators, developments of an eye, transkriptsionny factors: Rax, Pax6, Vsx2, Six3etc. result in various anomalies to developments of eyes, such as an anaftalmiya, колобома, a cataract, etc.
Objective of this researchis the existential characteristic of genes of Rax and Chx in families a course of a morphogenesis of a retina of hens
Genes of Rax and Chx families play important role in a retina morphogenesis. The structure of these genes vertebral and invertebrate is quite conservative. The quantity of genes varies at different types, however the pattern of an expression of these genes is similar, but isn't identical. Now with the help Ptsr – the analysis with specific praymer us are identified транскрипты Chxin a retina of embryos of hens from 4 to 18 days of development. Level an expression tends to increase from 4 to 8 days, at the subsequent stages doesn't change. The expression of cRaxLis detected at all studied stages.
Immunohistochemistry method the expression of other member of family Rax, cRax, is observed at early stages of development of embryos of hens: 3 - 5th days - in both neyroblastichesky layers to a retina, by 6-8 days - generally in gangliozny cages, and signal strength increases to a peripheral part of a retina and tsiliarny folds. The obtained data indicate participation of the studied genes in neurogenesis to a retina.
Further researches of genes of Rax and Chx families are necessary for understanding of genetic control of formation of a retina of vertebrata and the reasons of emergence of anomalies of development of an eye. Со
