Elaboration of internal inhibition and increase of activity of GABA neurotransmitter system as factors of weakening of a freezing and disinhibition of research behavior

Two series of experiments on rats of line of Wistar in the conditions of free behavior are carried out: 1) at development of a conditioned reflex of passive avoidance (the rat was trained not to run in a dark compartment of a cage as here she received electric stimulation of pads) and 2) at development of a conditional inhibition and a defensive reflex on light flashes at a reinforcement their electroskin stimulation of the hind extremity. In both series of experiments checked action of phenibut – a nonspecific agonist of an inhibitory mediator – gamma aminobutiric acid (GABA) on training. It is revealed, as elaboration of internal inhibition, and phenibut– more, than at a half of experimental animals cause weakening of a freezing, analog alarm and fear in the people arising in a dangerous situation, and a disinhibition of research behavior. Results of the experiments at the accounting of data of literature allow to assume that both factors: elaboration of internal inhibition and phenibut – a derivative of GABA inhibit the freezing owing to increase of level of activity of GABA neurotransmitter system of a brain. Our data will be coordinated with conclusions of the literary review that the condition of a depression at people considerably is defined by decrease in work of GABA neurotransmitter system (Luscher, B. Shen, Q. and Sahir N. 2011). Registration of activity of neurons of a cerebral cortex when training showed that the phenibut amplifies both inhibitory hyperpolarizing processes, and activation components of neuronal reactions to applied stimuli (Shulgina, 2005). Therefore, agonists of GAMK- receptors can be a basis for development of anxiolytics and antidepressants. The results received in our work show that phenibut – the nonspecific agonist of GABA-receptors which strengthens both inhibition and activation components of reactions of cortex neurons and provides replacement of a condition of alarm and fear with research activity, is one of such anxiolytics and, it may be, antidepressants.
