In the last years the testosterone-based drugs as treatment of androgen deficit in men have become widely used. Because of publications about androgenic effect on the mucosa structure and microflora of the colon are lacking, the aim of the investigation has been to study the influence of hyperandrogenemia on cell population of mucous membrane and on the gut organisms of the colon.

Sexually matured males Wistar rats, weight 220-240 gr. were used in the study. Hyperandrogenemia was produced by injection of the Omnadren 250® solution (165mg/kg). Animals of this group were removed on the 7th an 21st day after the injection. In order to define the levels of total testosterone and endotoxin the blood was taken from necked veins of Zoletil-anaesthetized animals. Level of total testosterone was measured in the serum using the ELISA method (“Adaltis” test-system). Endotoxin level was measured using LAL testing. For studying of luminal microflora received rectum contents. The colon was the study objects of the cell population of mucous membrane. The histological sections painted hematoxylin and eosin. Serotonin releasing by EC1-cells were revealed using Grimelius argyrophil reaction. The received data were statistically estimated.

After a single injection of Omnadren 250®, with comparison to the control group, the testosterone level in the blood serum was elevated 7 times on the 7 day, and on the 21 day it exceeded the one of the control group 5 times. Comparing with the control group, on 21 day there was found a certain decrease in

Volume fraction of the goblet cells to a half, the enterocytes were increased 20 %, the number of mitosis was increased 2,5 times, , the number of intraepithelial lymphocytes increased 2,5 times. The tendency for number reduction of the EC-1 cells was found. The epithelial-stromal relations had no changes. In the infiltrate the lymphocytes and granulocytes number increased 1,5 times. It was found a significant individual variability of the luminal microbiota in the examined data. Nevertheless it was found the tendency for decreasing in amount of the enterobacteria Lac+ and Lac -, the enterococci E. Faecalis and E. faecium, Lactobacteria, which reflects the onset of disbiotic condition. The blood serum endotoxin level also decreased. Thus, the Omnadren 250 injection in high doses leads to increasing in proliferative processes, shifting of the epithelial cells differentiation towards the limbic enterocytes , with decreasing of the EC1-cells and the volume fraction of the goblet cells .
